A morphometric and molecular study of Anastrepha pickeli Lima (Diptera: Tephritidae) Neotropical Entomology
Bomfim,ZV; Lima,KM; Silva,JG; Costa,MA; Zucchi,RA.
This study investigated the level of morphometric and genetic variability among populations of Anastrepha pickeli Lima from several localities in Brazil, one locality in Bolivia and one in Paraguay. Traditional and geometric morphometric analyses were used, as well as sequencing of a fragment of the cytochrome oxidase gene (COI). Six variables were measured from the aculeus for traditional morphometric analysis and 14 landmarks from the right wing were used for geometric analysis, using 10 specimes/population. The aculeus tip length, aculeus width at the end of the cloaca opening, and the serrate part length contributed with 62.7% for grouping. According to the results from traditional morphometry, there was no significant difference, but the multivariate...

A synopsis of the saddle fungi (Helvella: Ascomycota) in Europe – species delimitation, taxonomy and typification Naturalis
Skrede, I.; Carlsen, T.; Schumacher, T..
Helvella is a widespread, speciose genus of large apothecial ascomycetes (Pezizomycete: Pezizales) that are found in terrestrial biomes of the Northern and Southern Hemispheres. This study represents a beginning on assessing species limits and applying correct names for Helvella species based on type material and specimens in the university herbaria (fungaria) of Copenhagen (C), Harvard (FH) and Oslo (O). We use morphology and phylogenetic evidence from four loci – heat shock protein 90 (hsp), translation elongation factor alpha (tef), RNA polymerase II (rpb2) and the nuclear large subunit ribosomal DNA (LSU) – to assess species boundaries in an expanded sample of Helvella specimens from Europe. We combine the morphological and phylogenetic information...

Characterization of actin genes in Bonamia ostreae and their application to phylogeny of the Haplosporidia ArchiMer
Lopez Flores, L; Suarez Santiago, V; Longet, D; Saulnier, Denis; Chollet, Bruno; Arzul, Isabelle.
Bonamia ostreae is a protozoan parasite that infects the European flat oyster Ostrea edulis, causing systemic infections and resulting in massive mortalities in populations of this valuable bivalve species. In this work, we have characterized B. ostreae actin genes and used their sequences for a phylogenetic analysis. Design of different primer sets was necessary to amplify the central coding region of actin genes of B. ostreae. Characterization of the sequences and their amplification in different samples demonstrated the presence of 2 intragenomic actin genes in B. ostreae, without any intron. The phylogenetic analysis placed B. ostreae in a clade with Minchinia tapetis, Minchinia teredinis and Haplosporidium costale as its closest relatives, and...

Contribution of WUSCHEL-related homeobox (WOX) genes to identify the phylogenetic relationships among Petunia species Genet. Mol. Biol.
Segatto,Ana Lúcia Anversa; Thompson,Claudia Elizabeth; Freitas,Loreta Brandão.
Abstract Developmental genes are believed to contribute to major changes during plant evolution, from infrageneric to higher levels. Due to their putative high sequence conservation, developmental genes are rarely used as molecular markers, and few studies including these sequences at low taxonomic levels exist. WUSCHEL-related homeobox genes (WOX) are transcription factors exclusively present in plants and are involved in developmental processes. In this study, we characterized the infrageneric genetic variation of Petunia WOX genes. We obtained phylogenetic relationships consistent with other phylogenies based on nuclear markers, but with higher statistical support, resolution in terminals, and compatibility with flower morphological changes.

Cyanobacterial nitrogenases: phylogenetic diversity, regulation and functional predictions Genet. Mol. Biol.
Esteves-Ferreira,Alberto A.; Cavalcanti,João Henrique Frota; Vaz,Marcelo Gomes Marçal Vieira; Alvarenga,Luna V.; Nunes-Nesi,Adriano; Araújo,Wagner L..
Abstract Cyanobacteria is a remarkable group of prokaryotic photosynthetic microorganisms, with several genera capable of fixing atmospheric nitrogen (N2) and presenting a wide range of morphologies. Although the nitrogenase complex is not present in all cyanobacterial taxa, it is spread across several cyanobacterial strains. The nitrogenase complex has also a high theoretical potential for biofuel production, since H2 is a by-product produced during N2 fixation. In this review we discuss the significance of a relatively wide variety of cell morphologies and metabolic strategies that allow spatial and temporal separation of N2 fixation from photosynthesis in cyanobacteria. Phylogenetic reconstructions based on 16S rRNA and nifD gene sequences shed light on...

Davalliaceae in Peninsular Malaysia, a preliminary study based on trnL-F region Naturalis
Maideen, H.; Milne, R.I.; Gibby, M..
A preliminary molecular analysis based on trnL-F region is presented for 17 taxa of Davalliaceae in Peninsular Malaysia. Maximum parsimony and Bayesian analysis were conducted on the dataset in order to establish a robust phylogenetic relationship between taxa. The results of analysis indicate incongruence with morphological classification. All genera of Davalliaceae in the study area are paraphyletic except Araiostegia which is represented by only a single species. In addition it partially agrees with recent phylogeny base on rbcL data.

DNA barcoding of a tropical anguillid eel, Anguilla bicolor (Actinopterygii: Anguilliformes), in Indo-Pacific region and notes on its population structure Rev. Bras. Zool.
Norarfan,Ahmad Fathi; Mokti,Siti Shazwani Azreena; Taha,Hussein; Amin,Muhamad; Ali,Muhamad; Arai,Takaomi.
ABSTRACT The tropical anguillid eel, Anguilla bicolor McCelland, 1844, includes two subspecies, Anguilla bicolor bicolor McCelland, 1844 and Anguilla bicolor pacifica Schmidt, 1928, and is distributed across the Indo-Pacific region. Although A. bicolor is widely distributed and recognized as an important fish resource in the Indo-Pacific region, few studies have been conducted on its genetic variation and population structure. DNA barcoding of A. bicolor specimens collected in the Indo-Pacific region was carried out in this study using mitochondrial cytochrome c oxidase subunit I. Anguilla bicolor was found to diverge genetically, which supported its classification into two different subspecies. In addition, our study showed that A. bicolor bicolor had two...

Genera of diaporthalean coelomycetes associated with leaf spots of tree hosts Naturalis
Crous, P.W.; Summerell, B.A.; Alfenas, A.C.; Edwards, J.; Pascoe, I.G.; Porter, I.J.; Groenewald, J.Z..
Four different genera of diaporthalean coelomycetous fungi associated with leaf spots of tree hosts are morphologically treated and phylogenetically compared based on the DNA sequence data of the large subunit nuclear ribosomal DNA gene (LSU) and the internal transcribed spacers and 5.8S rRNA gene of the nrDNA operon. These include two new Australian genera, namely Auratiopycnidiella, proposed for a leaf spotting fungus occurring on Tristaniopsis laurina in New South Wales, and Disculoides, proposed for two species occurring on leaf spots of Eucalyptus leaves in Victoria. Two new species are described in Aurantiosacculus, a hitherto monotypic genus associated with leaf spots of Eucalyptus in Australia, namely A. acutatus on E. viminalis, and A....

Host specificity and phylogenetic relationships among Atlantic Ovulidae (Mollusca: Gastropoda) Naturalis
Reijnen, B.T.; Hoeksema, B.W.; Gittenberger, E..
Ovulid gastropods and their octocoral hosts were collected along the leeward coast of Curaçao, Netherlands Antilles. New molecular data of Caribbean and a single Atlantic species were combined with comparable data of Indo-Pacific Ovulidae and a single East-Pacific species from GenBank. Based on two DNA markers, viz. CO-I and 16S, the phylogenetic relationships among all ovulid species of which these data are available are reconstructed. The provisional results suggest a dichotomy between the Atlantic and the Indo-Pacific taxa. Fully grown Simnialena uniplicata closely resembles juvenile Cyphoma gibbosum conchologically. Cymbovula acicularis and C. bahamaensis might be synonyms. The assignments of Caribbean host species for Cyphoma gibbosum, C....

ITS-rDNA phylogeny of Colletotrichum spp. causal agent of apple Glomerella leaf spot Ciência Rural
Giaretta,Diorvania Ribeiro; Bogo,Amauri; Coelho,Cileide Maria Medeiros; Guidolin,Altamir Frederico; Dantas,Adriana Cibele de Mesquita; Gomes,Eliane Aparecida.
Several diseases have affected apple production, among them there is Glomerella leaf spot (GLS) caused by Colletotrichum spp. The first report of this disease in apple was in plants nearby citrus orchards in São Paulo State, Brazil. The origin of this disease is still not clear, and studies based on the molecular phylogeny could relate the organisms evolutionarily and characterize possible mechanisms of divergent evolution. The amplification of 5.8S-ITS (Internal Transcribed Spacer) of rDNA of 51 pathogenic Colletotrichum spp. isolates from apples, pineapple guava and citrus produced one fragment of approximately 600 bases pairs (bp) for all the isolates analyzed. The amplified fragments were cleaved with restriction enzymes, and fragments from 90 to 500bp...

Juglanconis gen. nov. on Juglandaceae, and the new family Juglanconidaceae (Diaporthales) Naturalis
Voglmayr, H.; Castlebury, L.A.; Jaklitsch, W.M..
Molecular phylogenetic analyses of ITS-LSU rDNA sequence data demonstrate that Melanconis species occurring on Juglandaceae are phylogenetically distinct from Melanconis s.str., and therefore the new genus Juglanconis is described. Morphologically, the genus Juglanconis differs from Melanconis by light to dark brown conidia with irregular verrucae on the inner surface of the conidial wall, while in Melanconis s.str. they are smooth. Juglanconis forms a separate clade not affiliated with a described family of Diaporthales, and the family Juglanconidaceae is introduced to accommodate it. Data of macro- and microscopic morphology and phylogenetic multilocus analyses of partial nuSSU-ITS-LSU rDNA, cal, his, ms204, rpb1, rpb2, tef1 and tub2 sequences revealed...

Meiofaunal cryptic species challenge species delimitation: the case of the Monocelis lineata (Platyhelminthes: Proseriata) species complex Naturalis
Scarpa, F.; Cossu, P.; Lai, T.; Sanna, D.; Curini-Galletti, M.; Casu, M..
Given the pending biodiversity crisis, species delimitation is a critically important task in conservation biology, but its efficacy based on single lines of evidence has been questioned as it may not accurately reflect species limits and relationships. Hence, the use of multiple lines of evidence has been portrayed as a means to overcome identification issues arising from gene/ species tree discordance, morphological convergence or recent adaptive radiations. Here, the integrative taxonomic approach has been used to address the study of the Monocelis lineata species complex. The taxonomic resolution of the complex is challenging, as the species lacks sclerotised copulatory structures, which as a rule of thumb aid identification in Proseriata. Eighteen...

Molecular Characterization of Novel Cryptosporidium Fish Genotypes in Edible Marine Fish ArchiMer
Certad, Gabriela; Zahedi, Alireza; Gantois, Nausicaa; Sawant, Manasi; Creusy, Colette; Duval, Erika; Benamrouz-vanneste, Sadia; Ryan, Una; Viscogliosi, Eric.
Current knowledge of Cryptosporidium species/genotypes in marine fish is limited. Following phylogenetic analysis at the 18S rDNA locus, a recent study identified six new genotypes of Cryptosporidium colonizing edible fish found in European seas. Of these, five grouped in a clade together (#Cryptofish 1–5) and one grouped separately (#Cryptofish 7). In the present study, after phylogenetic analyses of #Cryptofish1, #Cryptofish2, #Cryptofish4, #Cryptofish5 and #Cryptofish7 at the actin locus, the presence of two major clades was confirmed. In addition, when possible, longer 18S amplicons were generated. In conclusion, the small genetic distances between these genotypes designated as a novel marine genotype I (#Cryptofish 1-5) suggest that they may be...
